SV40 in vaccines is not a novelty but have been used in vaccines to create cancers for decades. Merck, Sharp and Dome isolated LS-c, 2ab/KP2 passed strain of rhesus monkey kidney cell culture with SV40 contaminant for the Polio Vaccines.

Controlled Co-assembly of Viral Nanoparticles of Simian Virus 40 with Inorganic Nanoparticles
Simian virus 40 (SV40) has been one of the viruses exploited as VNPs for nanobiotechnology development. SV40 VNPs are assembled as cage-like structures from the major capsid protein VP1. Assembly is the principal way to integrate VNPs with chemically synthesized nanomaterials. A variety of inorganic nanoparticles (NPs) of different components, surface coatings, shapes, and sizes have been encapsulated into SV40 VNPs through controllable assembly. NPs can also be loaded onto the outer surface by rationally tuning the interfacial interactions between SV40 VNPs and NPs. Biomineralization is another route for combining VNPs with inorganic NPs, by which noble metal plasmonic NPs with tunable sizes and components have been produced in SV40 VNPs. By controllably assembling these bio-nanohybrid structures, different functionalities have been integrated for nanophotonic studies and bioimaging. These studies have laid a foundation for the development of multifunctional bio-nanomaterials and devices based on the SV40 VNP platform.
SV40 platform also has been used to create Quantum Dots, which are tiny semiconductor particles with unique optical and electronic properties.
Quantum dot-induced viral capsid assembling in dissociation buffer
Viruses encapsulating inorganic nanoparticles are a novel type of nanostructure with applications in biomedicine and biosensors. However, the encapsulation and assembly mechanisms of these hybridized virus-based nanoparticles (VNPs) are still unknown. In this article, it was found that quantum dots (QDs) can induce simian virus 40 (SV40) capsid assembly in dissociation buffer, where viral capsids should be disassembled. The analysis of the transmission electron microscope, dynamic light scattering, sucrose density gradient centrifugation, and cryo-electron microscopy single particle reconstruction experimental results showed that the SV40 major capsid protein 1 (VP1) can be assembled into ≈25 nm capsids in the dissociation buffer when QDs are present and that the QDs are encapsulated in the SV40 capsids. Moreover, it was determined that there is a strong affinity between QDs and the SV40 VP1 proteins (KD = 2.19E-10 M), which should play an important role in QD encapsulation in the SV40 viral capsids. This study provides a new understanding of the assembly mechanism of SV40 virus-based nanoparticles with QDs, which may help in the design and construction of other similar virus-based nanoparticles.

Encapsulation of gold nanoparticles by simian virus 40 capsids
Viral capsid-nanoparticle hybrid structures constitute a new type of nanoarchitecture that can be used for various applications. We previously constructed a hybrid structure comprising quantum dots encapsulated by simian virus 40 (SV40) capsids for imaging viral infection pathways. Here, gold nanoparticles (AuNPs) are encapsulated into SV40 capsids and the effect of particle size and surface ligands (i.e. mPEG and DNA) on AuNP encapsulation is studied. Particle size and surface decoration play complex roles in AuNP encapsulation by SV40 capsids. AuNPs ≥15 nm (when coated with mPEG750 rather than mPEG2000), or ≥10 nm (when coated with 10T or 50T DNA) can be encapsulated. Encapsulation efficiency increased as the size of the AuNPs increased from 10 to 30 nm. In addition, the electrostatic interactions derived from negatively charged DNA ligands on the AuNP surfaces promote encapsulation when the AuNPs have a small diameter (i.e. 10 nm and 15 nm). Moreover, the SV40 capsid is able to carry mPEG750-modified 15-nm AuNPs into living Vero cells, whereas the mPEG750-modified 15-nm AuNPs alone cannot enter cells. These results will improve our understanding of the mechanisms underlying nanoparticle encapsulation in SV40 capsids and enable the construction of new functional hybrid nanostructures for cargo delivery.

Nucleotide and DNA coordinated lanthanides: From fundamentals to applications
Lanthanides are very important for their unique catalytic, optical and magnetic properties. Various ligands have been synthesized to coordinate with lanthanides so that their properties can be better controlled enabling various analytical, materials and biomedical applications. DNA is an anionic polymer made of four types of nucleotides. With high stability, low cost, programmability, and molecular recognition and catalytic functions, DNA has become a very popular material in the past few decades. In recent years, both nucleotides and DNA have been explored as ligands for lanthanides. In this article, fundamental interactions between nucleotides and DNA with lanthanides are reviewed in terms of binding sites and thermodynamics. Further, coordination polymers formed by mixing nucleotides and lanthanides are discussed, which can form nanoparticles, fibers and hydrogels with applications for enzyme encapsulation, imaging and biosensors.
Encapsulation of Inorganic Nanomaterials inside Virus-Based Nanoparticles for Bioimaging
Virus-based nanoparticles (VNPs) can serve as containers for inorganic nanomaterials with excellent physical and chemical properties. Incorporation of nanomaterials inside the inner cavity of VNPs has opened up lots of possibilities for imaging applications in the field of biology and medicine. Encapsulation of inorganic nanoparticles (NPs) in VNPs can achieve the labeling of VNPs with nanoprobes and maintain the original outer surface features of VNPs at the same time. In return, VNPs enhance the stability and biocompatibility of the inorganic cargoes. This review briefly summarizes the current typical strategies to encapsulate inorganic nanomaterials in VNPs, i.e. mineralization and self-assembly, as well as the applications of these hybrid nanostructures in the field of bioimaging, including in vitro and in vivo fluorescence imaging, magnetic resonance imaging, and theranostics. Nanophotonic studies based on the VNP platform are also discussed. We anticipate that this field will continue to flourish, with new exciting opportunities stemming from advancements in the rational design of VNPs, the development of excellent inorganic nanomaterials, the integration of multiple functionalities, and the regulation of nano-bio interfacial interactions.
RNA encapsidation by SV40-derived nanoparticles follows a rapid two-state mechanism
Viruses are evolved examples of self-assembled structures that are remarkably sophisticated biological machines. This ability to self-assemble can be harnessed for nano- and bio-technology.

Replication-defective (RD) recombinant simian virus 40 (SV40)-based gene delivery vectors hold a great potential for clinical applications because of their presumed non-immunogenicity and capacity to induce immune tolerance to the transgene products in humans. However, the clinical use of SV40 vectors has been hampered by the lack of a packaging cell line that produces replication-competent (RC) free SV40 particles in the vector production process. To solve this problem, we have adapted the current SV40 vector genome used for the production of vector particles and generated a novel Vero-based packaging cell line named SuperVero that exclusively expresses the SV40 large T antigen. SuperVero cells produce similar numbers of SV40 vector particles compared to the currently used packaging cell lines, albeit in the absence of contaminating RC SV40 particles. Our unique SV40 vector platform named SVac paves the way to clinically test a whole new generation of SV40-based therapeutics for a broad range of important diseases.
